Resumo(s): | O desenvolvimento de aplicações e sistemas de libertação controlada emerge como uma área
bastante promissora, surgindo nesse âmbito a aposta na utilização de polímeros responsivos
capazes de responder às necessidades específicas de cada pessoa e de cada situação.
A presente dissertação foi desenvolvida no âmbito do estágio realizado no CeNTI – Centro de
Nanotecnologia e Materiais Técnicos, Funcionais e Inteligentes. Dentro da área de investigação
dos materiais funcionais da empresa, surgiu a oportunidade de desenvolver um estudo sobre
sistemas de libertação controlada baseados em polímeros pH-responsivos. Para tal, foi
inicialmente estudado um procedimento de síntese para obter nanopartículas a partir de polímeros
pH-responsivos disponíveis comercialmente (com e sem incorporação de fármaco), em seguida
essas partículas foram sujeitas a técnicas de caracterização, e por fim, realizaram-se estudos de
libertação do fármaco incorporado com as nanopartículas sintetizadas.
Na otimização do processo de síntese destacou-se uma melhoria no tamanho e estabilidade das
nanopartículas sintetizadas recorrendo à utilização de Span® 85 em vez de álcool polivinílico e de
velocidades de agitação mais elevadas durante a síntese.
Na caracterização das nanopartículas sintetizadas foi possível constatar que as mesmas possuíam
tamanhos dentro dos limites aceitáveis para sistemas de libertação controlada e valores de
potencial zeta indicadores da sua elevada estabilidade. No caso particular das nanopartículas com
incorporação de ibuprofeno, demonstrou-se, através das análises de caracterização, que
efetivamente o ibuprofeno havia sido incorporado, com exceção das nanopartículas de Eudragit®
L100-55, nas quais não foi possível a incorporação do fármaco.
Através dos estudos de libertação com as nanopartículas de Eudragit® L100-55 obtidas pré- e
pós-liofilização, demonstrou-se que não ocorreu libertação já que não existia ibuprofeno
incorporado. Nos estudos com Eudragit® E PO demonstrou-se que, para as nanopartículas obtidas
pré-liofilização, com o aumento de ibuprofeno nas partículas ocorreu um aumento da velocidade
de libertação, sendo que a velocidade foi sempre maior para o estudo a pH 5,5. No caso das
nanopartículas obtidas pós-liofilização, com o aumento de ibuprofeno nas partículas ocorreu a
diminuição da velocidade da libertação, sendo que a velocidade foi maior para pH 6,8. The development of applications and drug delivery systems emerge as a quite promising area, arising within this context the potential use of responsive polymers capable of addressing the specific needs of each person and situation. The present dissertation was developed under the scope of the internship performed at CeNTI - Centre for Nanotechnology and Smart Materials. Within the functional materials research area of the company, there was an opportunity to conduct a study about drug delivery systems based on pH-responsive polymers. Therefore, it was initially studied a synthesis procedure to obtain nanoparticles from pH responsive polymers commercially available (with or without drugs incorporated), then those particles were characterized using a number of techniques, and, at last drug release studies with the synthesized nanoparticles were developed. In the optimization of the synthesis process, the synthesized nanoparticles size and stability was improved through the use of Span® 85 instead of poly(vinyl alcohol) and higher agitation velocities during the synthesis. Throughout the characterization of the synthesized nanoparticles it was found that they possess sizes within the acceptable limits for drug delivery systems and exhibited zeta potential values that indicate their high stability. Regarding the nanoparticles loaded with ibuprofen it was shown, through characterization techniques, that the ibuprofen was indeed incorporated in the nanoparticles, with the exception of the Eudragit® L100-55 nanoparticles, in which the incorporation has not been possible. From the release studies conducted with the Eudragit® L100-55 nanoparticles obtained before and after the freeze-drying procedure, it was shown that the release does not occur since there was no ibuprofen incorporated. In the studies with Eudragit® E PO, it was found that, regarding the nanoparticles obtained before freeze-drying, with the increase of ibuprofen in the particles there was an increase in the release velocity, wherein the velocity was always higher for the study at pH 5,5. On the other hand, when it comes to the case of the nanoparticles obtained after freeze-drying, with the increase of ibuprofen in the particles, it occurred the decrease of the release velocity, wherein the velocity was higher for the study at the pH 6,8. |
