Resumo(s): | As terapias atuais para o tratamento de doenças relativas ao sistema nervoso apresentam muitas
limitações, traduzindo-se em resultados pouco eficientes e duradouros. Uma estratégia que pode ser
revolucionária é a engenharia de tecidos no contexto neuronal, uma vez que esta é capaz de promover
a regeneração e a proliferação de células nervosas. Com este objetivo os materiais piezoelétricos
demonstram-se úteis, pois estes permitem a estimulação de células de forma localizada e pouco invasiva.
Nesta dissertação utilizou-se filmes de poli(fluoreto de vinilideno) (PVDF) polarizados e não
polarizados e foram produzidas fibras de PVDF orientadas e não orientadas. Todos estes materiais foram
caracterizados a nível morfológico, físico-químico e mecânico. Também foi avaliado o efeito destes
materiais no comportamento de astrócitos, células fundamentais para o bom funcionamento do sistema
nervoso e que estão associados à proteção e suporte do mesmo. Para esta análise foram realizados
ensaios de proliferação celular em condições estáticas e dinâmicas.
Com as morfológicas foi possível verificar que as fibras apresentam uma maior rugosidade do
que os filmes, e que a técnica aplicada para obter diferentes orientações nas fibras não alterou
significativamente o diâmetro das mesmas. A avaliação físico-química demonstrou que a fração de fase
β das fibras é superior à dos filmes, sendo esta superior a 90%. As análises mecânicas demonstraram
que os filmes de PVDF possuem módulos de Young elevados quando comparados com as fibras.
Também foi verificado que os módulos de Young variam com a orientação das fibras, sendo mais elevado
nas fibras orientadas.
Os ensaios celulares demonstraram que as células aderem e proliferam em todas as amostras
de PVDF, sendo que nas fibras os melhores resultados foram obtidos nas orientadas. Em relação à carga
dos filmes de PVDF, a carga positiva parece favorecer o crescimento dos astrócitos. Também se
demonstrou que a aplicação de estímulos mecânicos promoveu a proliferação dos astrócitos em todas
as amostras. Importa ainda notar que os resultados em relação ao efeito da poli-d-lisina são ambíguos já
que nos filmes esta diminui a proliferação dos astrócitos, mas nas fibras aumenta. Comprovou-se assim
que os materiais desenvolvidos e os estímulos fornecidos por eles foram capazes de influenciar a
proliferação de astrócitos, demonstrando a sua potencialidade para novas abordagens em engenharia do
tecido neuronal. Currently existing therapies in the treatment of diseases related to the nervous system have many limitations, resulting in inefficient and fleeting results. One strategy that could be revolutionary is tissue engineering in the neuronal context, as it is able to promote the regeneration and proliferation of nerve cells. To this end, piezoelectric materials have been shown to be useful, as they allow the stimulation of cells in a localized and minimally invasive way. In this dissertation, commercial polarized and non-polarized poly(vinylidene fluoride) (PVDF) films were used and PVDF fibers with different orientations were produced. All these materials were characterized at the morphological, physicochemical, and mechanical levels. The effect of these materials on the behavior of astrocytes, cells that are fundamental for the proper functioning of the nervous system and that are associated with its protection and support, was also evaluated. For this analysis, cell proliferation assays were performed under static and dynamic conditions. From the morphological analyses, we were able to verify that the fibers have more roughness than the films, and that the technique applied to obtain different orientations in the fibers did not significantly change their diameter. The physicochemical evaluation showed that the β phase fraction of the fibers is higher than that of the films reaching 90%. Mechanical analyses showed that commercial films have high Young's modulus when compared to fibers. It was also found that Young's modulus change with the fiber orientation, being higher in the oriented ones. The cellular assays showed that the cells respond well to PVDF in the different morphologies used, and in the fibers the best results were obtained in the oriented ones. Regarding the charge of the films, the positive charge seems to favor the growth of astrocytes. Through this assay it was also demonstrated that the application of mechanical stimuli promoted the proliferation of astrocytes in all samples. It is also important to note that the results regarding the effect of poly-d-lysine are ambiguous since in the films it decreased the proliferation of astrocytes, but in the fibers it increased. Thus, it was proven that the developed materials and the stimuli provided by them were able to influence the proliferation of astrocytes, demonstrating a new possibility for neural tissue engineering. |
