Resumo(s): | Candida albicans é um fungo comensal, presente na flora microbiana do hospedeiro, mas com
capacidade de colonizar outros microambientes e tornar-se patogénico. A patogenicidade está associada
à sua flexibilidade metabólica, resposta a stresses provocados pelo hospedeiro, resistência a fagocitose,
rápida transição morfológica e a outros fatores de virulência, que fazem de C. albicans uma das principais
causas de infeções fúngicas. A flexibilidade metabólica permite que C. albicans utilize diversas fontes de
carbono, entre as quais os ácidos carboxílicos. Para isso, este microrganismo possui transportadores
transmembranares, capazes de fazer o importe destes substratos. Os transportadores Jen1 e Jen2 de
C. albicans foram identificadas como permeases de monocarboxilatos e dicarboxilatos, respetivamente.
As proteínas da membrana plasmática Atos(Acetate Transporter Orthologs) da família AceTr são também
potenciais transportadores de ácidos carboxílicos em C. albicans. Em Saccharomyces cerevisiae o
transportador Ato1 foi descrito como transportador de acetato. Em C. albicans existem dez genes ATO,
e entre eles, dois são possivelmente não-funcionais.
O objetivo deste trabalho foi estudar os transportadores Ato e Jen de S. cerevisiae e de C. albicans
por expressão homóloga e heteróloga e também por ensaios fenotípicos com estirpes mutantes de C.
albicans. Para tal, foram construídos 11 novos plasmídeos por Gap-Repair contendo os genes ATO e JEN
de S. cerevisiae e C. albicans, sob o controlo do promotor GPD. Por último, foram feitas análises in silico
de todos os Atos de C. albicans e ainda previsões do raio do poro dos transportadores Ato6, Ato7 e Ato8.
Os resultados mostraram que o Ato1 e o Ato3 de S. cerevisiae desempenham um papel
importante na utilização de ácidos carboxílicos. Em C. albicans, o Ato1 parece ser relevante na
assimilação de monocarboxilatos. Os mutantes simples de C. albicans nos restantes Atos não
apresentaram variações fenotípicas, no entanto, poderão mesmo assim estar envolvidos no transporte e
utilização de carboxilatos. Das análises in silico, conclui-se que as regiões NPAPLGL(M/S), SYG(X)FW e
FLY estão conservadas na maioria dos Atos. As previsões dos raios dos poros do Ato6, Ato7 e Ato8
mostraram que a região do motivo FLY é o local de maior constrição e que o raio reduzido desta região
poderá funcionar como um mecanismo de seleção de substratos. Por último, estes Atos apresentaram
uma constrição inferior quando comparados com o Ato1 de S. cerevisiae, o que pode indicar que estes
transportam substratos maiores. Candida albicans is a commensal fungus, present in the microbial flora of the host, but capable of colonizing other microenvironments and becoming pathogenic. Pathogenicity is associated with its metabolic flexibility, response to host stresses, resistance to phagocytosis, rapid morphological transition and other virulence factors, which make C. albicans one of the main causes of fungal infections. Metabolic flexibility allows C. albicans to use different carbon sources, including carboxylic acids. For this, this microorganism has transmembrane transporters, capable of importing these substrates. The C. albicans Jen1 and Jen2 transporters have been identified as monocarboxylate and dicarboxylate permeases, respectively. The plasma membrane proteins Atos (Acetate Transporter Orthologs) of the AceTr family are also putative carboxylic acid transporters in C. albicans. In Saccharomyces cerevisiae the Ato1 transporter has been described as an acetate transporter. In C. albicans there are ten ATO genes, and among them, two are possibly non-functional. The objective of this work was to study the Ato and Jen transporters of S. cerevisiae and C. albicans by homologous and heterologous expression and also by phenotypic assays with mutant strains of C. albicans. For this purpose, 11 new plasmids were constructed by Gap-Repair containing the ATO and JEN genes from S. cerevisiae and C. albicans, under the control of the GPD promoter. Finally, in silico analyses of all Atos of C. albicans were performed, as well as predictions of the pore radius of the transporters Ato6, Ato7 and Ato8. The results showed that S. cerevisiae Ato1 and Ato3 play an important role in the utilization of carboxylic acids. In C. albicans, Ato1 appears to be relevant in the assimilation of monocarboxylates. The single mutants of C. albicans in the remaining Atos did not show phenotypic variations, however, they could still be involved in the transport and use of carboxylates. From the in silico analyses, it is concluded that the NPAPLGL(M/S), SYG(X)FW and FLY regions are conserved in most Atos. The predictions of the pore radii of Ato6, Ato7 and Ato8 showed that the region of the FLY motif is the site of greatest constriction and that the reduced radius of this region could function as a substrate selection mechanism. Finally, these Atos showed a lower constriction when compared to Ato1 of S. cerevisiae, which may indicate that they transport larger substrates. |
