Resumo(s): | A glicosilação é uma modificação molecular, mediada por enzimas, crucial em muitos processos
biológicos. Alterações no processo de glicosilação resultam na expressão aberrante de glicanos e,
consequentemente podem influenciar o normal funcionamento da célula. De facto, estudos
mostram que a transformação maligna das células é acompanhada pela desregulação da
expressão de glicosiltransferases, enzimas que adicionam os vários açúcares, levando à
biossíntese de estruturas aberrantes de glicanos associadas ao cancro. Uma das alterações mais
comuns consiste na expressão dos O-glicanos truncados Tn (GalNAcα-O-Ser/Thr) e Sialyl-Tn (STn Neu5Acα-2,6GalNAcα-O-Ser/Thr). O antigénio STn é expresso em vários tipos de cancro, estando
associado a uma maior agressividade dos tumores e a um pior prognóstico dos doentes. Assim, o
glicano truncado STn apresenta-se como um importante alvo terapêutico. Estudos recentes
mostraram a eficácia dos análogos fluorados de monossacarídos (F-MAs) em inibir a biossíntese
de vários glicanos associados a cancro. Assim, o uso de F-MAs representa uma estratégia
inteligente para inibir a biossíntese do glicano STn.
Devido à falta de modelos celulares para estudar O-glicanos truncados, o principal objetivo desta
tese foi estabelecer modelos celulares, recorrendo à técnica de edição do genoma por
CRISPR/Cas9, que expressassem os glicanos Tn e STn. Após estabelecimento das linhas celulares
fomos estudar a capacidade do análogo GalNAc fluorado em afetar a biossíntese desses mesmos
glicanos. Neste trabalho foram selecionadas 3 linhas celulares derivadas de cancros do trato
gastrointestinal, de acordo com o nível de expressão da enzima ST6GalNAc-I, enzima que adiciona
o acido siálico ao antigénio Tn formando STn; nas quais foi feito posteriormente o KnockOut (KO)
da enzima C1GalT1, enzima que faz a extensão de O-glicanos. O KO foi validado por técnicas de
analise genómica por polimorfismo de comprimento de fragmentos de DNA (RFLP) e sequenciação
de sanger dos clones selecionados. Por Western Blot e imunofluorescência foi validada a
expressão dos antigénios Tn e STn. Estes modelos foram usados para testar 4F-GalNAc que
demonstrou ser eficaz no bloqueio in vitro da biossíntese de Tn e STn em dois dos modelos
celulares estabelecidos. Assim, nesta tese foram estabelecidos com sucesso três modelos
celulares de cancro que expressam os antigénios Tn e STn, modelos estes que poderão ser
utilizados em vários estudos na área da glicobiologia em cancro. Além disso, este trabalho mostra
o potencial do análogo fluorado do açúcar GalNAc como um inibidor da biossíntese de O-glicanos. Glycosylation is a molecular modification, enzymatically mediated, essential in many biological processes. Changes in the glycosylation process result in the aberrant expression of glycans and consequently, can influence the function of the cell. In fact, studies have been showing that malignant transformation of cells is accompanied by the dysregulation of glycosyltransferases expression, the enzymes that catalyze sugar’s transfer, leading to the biosynthesis of aberrant cancer-associated glycan structures. One of the most common alterations is the expression of the truncated O-glycans Tn (GalNAcα-O-Ser/Thr) and Sialyl-Tn (STn-Neu5Acα-2,6GalNAcα-O-Ser/Thr). The STn antigen is expressed in various type of tumors and its expression is associated with increase tumors’ aggressiveness and poor patients’ prognosis. Thus, the truncated O-glycan STn is an important therapeutic target. Recent studies have shown the efficacy of fluorinated monosaccharide (F-Mas) analogs in the inhibition of various cancer-associated glycans both in vitro and in vivo. Therefore, the use of F-MAs represents a clever strategy to impair the biosynthesis of STn, opening opportunities for translational studies. Due to the lack of cellular models to study truncated O-glycans, the main objective of this thesis was the development of glycoengineered cellular models, using CRISPR/Cas9 technology, to express Tn and STn antigens. After establishing the glycoengineered cellular models, we studied the capacity of fluorinated GalNAc in impairing STn biosynthesis. Three gastrointestinal cancer cell lines were selected according to the expression of ST6GALNAC1, the enzyme that catalyzes the addition of sialic acid to Tn antigen forming STn, and a KnockOut (KO) of the C1GalT1, an enzyme that extends O-Glycans enzyme, was performed. The KO was confirmed by genomic analyses using restriction fragment length polymorphism (RFLP) and sanger sequencing of the selected clones. By Western Blot and immunofluorescence, we validated the expression of Tn and STn antigens. These models were used to assess the capacity of the fluorinated monosaccharide GalNAc (4F-GalNAc) as an effective inhibitor of the biosynthesis of Tn and STn in vitro in two of the established cellular models. Altogether, in this thesis we successfully established three models of gastrointestinal cancer cell lines that express Tn and STn antigens. These models will open the opportunity to perform numerous studies in the glycobiology in cancer field. Furthermore, this work shows the potential of the fluorinated GalNAc as novel inhibitor of the biosynthesis of O-glycans. |
