Resumo(s): | A flavescência dourada é uma doença da videira, causada pelo fitoplasma Candidatus phytoplasma vitis,
propagado pelo vetor Scaphoideus titanus. Atualmente encontra-se disseminada pela maioria das regiões
vitivinícolas da Europa. Em Portugal foi detetada pela primeira vez em 2007 na Região Demarcada dos “Vinhos
Verdes” (DOC VV). Os sintomas mais visíveis da infeção consistem no atraso ou ausência do abrolhamento,
crescimento reduzido e redução da quantidade dos gomos florais e consequente elevada perda de produtividade.
Diferentes estudos apontam para alterações substanciais no metabolismo primário e secundário em videiras
infetadas, incluindo a redução das clorofilas e a consequente diminuição da taxa fotossintética, acumulação nas
folhas de açúcares e de compostos fenólicos, no entanto, os mecanismos moleculares envolvidos nestas alterações
metabólicas ainda são pouco conhecidos. Este estudo teve como principal objectivo a avaliação dos efeitos da
flavescência dourada em folhas de videira ao nível molecular e bioquímico, nomeadamente i) no metabolismo dos
principais açúcares e expressão de genes transportadores envolvidos no seu efluxo, ii) na síntese de pigmentos
fotossintéticos como as clorofilas a, b e carotenoides totais e expressão de genes envolvidos na sua síntese e iii)
em compostos-chave do metabolismo secundário e expressão de genes envolvidos nas principais vias da sua
biossíntese. Para o efeito, foi selecionada a variedade Loureiro de uma vinha comercial localizada na região dos
Vinhos Verdes, com plantas previamente diagnosticadas com infeção por FD. Foram colhidas folhas com sintomas
aparentes de infeção e folhas saudáveis (controlo) em dois períodos do desenvolvimento vegetativo (E-L 33 e E-L
38). Testes de detecção da doença por PCR em tempo real permitiram confirmar a infeção por FD apenas nas
folhas de videiras sintomáticas. Nestas folhas, verificou-se um forte decréscimo do conteúdo em clorofilas a e b
bem como do número de transcritos dos genes VvGGPPS11, VvGGR e VvCHLSYN envolvidos na sua biossíntese.
Em sentido oposto, folhas infetadas acumularam maior quantidade de açúcares redutores e sacarose, e foi
observada uma redução da expressão de genes da membrana plastidial envolvidos na importação/exportação de
açúcares incluindo o VvSWEET1, VvpGT1 e VvpGT2, VvGPT, VvMEX e gene VvNTT1 que codifica o antiportador de
ATP com ADP da membrana plastidial. Por outro lado, em resposta à infeção, ocorreu um aumento da
concentração de fenólicos totais, ácidos hidroxicinámicos flavonóis e flavan-3-ols, particularmente em folhas de
plantas na fase madura. No entanto, a expressão de alguns genes chave envolvidos na síntese destes metabolitos,
como o VvPAL e VvSTS, não sofreu alterações significativas. Foi ainda observado que a redução das clorofilas
poderá ser controlada a nível transcricional. O estudo bioquímico e molecular das alterações induzidas pelo
fitoplasma da flavescência dourada permitiu compreender mais detalhadamente a redução do conteúdo em
clorofilas e consequente o amarelecimento das folhas de videiras da variedade Loureiro, bem como alguns dos
mecanismos moleculares de resposta à infeção. Flavescence dorée is a vine disease, caused by the phytoplasma Candidatus phytoplasma vitis, propagated by the vector Scaphoideus titanus. It’s now widespread in most wine regions in Europe. In Portugal, it was detected in 2007 in the Demarcated Region of “Vinhos Verdes” (DOC VV). The most visible symptoms of the infection consist of delayed or absent budbreak, reduced growth and reduced quantity of floral buds and consequent high loss of production. Different studies point to substantial changes in primary and secondary metabolism in infected vines, including the reduction of chlorophylls and the consequent decrease in photosynthetic rate, accumulation of sugars and phenolic compounds in the leaves, however, the molecular mechanisms involved in these metabolic changes are still poorly studied. The main objective of this study was the evaluation of the effects of FD on vine leaves at the molecular and biochemical level, namely i) on the metabolism of the main sugars and expression of transporter genes involved in their efflux, ii) on the synthesis of photosynthetic pigments such as chlorophylls a, b and total carotenoids and expression of genes involved in their synthesis, and iii) in key compounds of secondary metabolism and expression of genes involved in the main pathways of their biosynthesis. For this purpose, the Loureiro variety from a commercial vineyard located in the DOC region “Vinhos Verdes” was selected, with plants previously diagnosed with FD infection. Leaves with apparent symptoms of infection and healthy leaves (control) were collected in two periods of vegetative development (E-L 33 and E-L 38). Tests for detection of the disease by real-time PCR confirmed the FD infection only in symptomatic grape leaves. In these leaves, there was a strong decrease in the content of chlorophylls a and b as well as in the number of transcripts of the VvGGPPS11, VvGGR and VvCHLSYN genes involved in their biosynthesis. Conversely, infected leaves accumulated a greater amount of reducing sugars and sucrose, and a reduction in the expression of plastidial membrane genes involved in the import / export of sugars was observed, including VvSWEET1, VvpGT1 and VvpGT2, VvGPT, VvMEX and VvNTT1 gene that encodes the antiporter ADP/ATP of the plastidial membrane. On the other hand, in response to the infection, there was an increase in the concentration of total phenolics, hydroxycinnamic acids, flavonols and flavan-3-ols, particularly in leaves of plants in the mature phase (E-L 38). However, the expression of some key genes involved in the synthesis of these metabolites, such as VvPAL and VvSTS, did not change significantly. It was also observed that the reduction of chlorophylls can be controlled at the transcriptional level. The biochemical and molecular study of the changes induced by the phytoplasma of FD allowed to understand in more detail the reduction in the content of chlorophylls and the consequent leaf yellowing of the cv. Loureiro, as well as some of the molecular mechanisms in response to infection. |
