Resumo(s): | As Moléculas intercalantes são usadas como agentes de deteção e
quantificação de ADN. Estas moléculas intercalam entre as cadeias duplas de
ADN, em solução aquosa, devido às interações hidrofóbicas existentes entre as
moléculas. Estas moléculas intercalantes são fluorescentes mas, quando
intercaladas, exibem um aumento significativo da sua intensidade de
fluorescência.
Este efeito não é observado quando a solução deixa de ser a solução
aquosa habitual, como por exemplo, uma solução composta por um líquido
iónico à temperatura ambiente (RTILs). O decréscimo da fluorescência dos
intercalantes poderá estar relacionada com o facto de estes não intercalarem
com o ADN, devido às fortes interações iónicas entre o solvente e a cadeia de
ADN, ou através de outros fenómenos envolvidos na interação entre as
moléculas estudadas.
Com este projeto pretendemos verificar se estamos perante um
fenómeno de exclusão por parte do Liquido Iónico, ou se a intercalação é
espontânea, estudando os mecanismos moleculares envolvidos intercalação na
cadeia de ADN e em que condição a interação entre ADN e intercaladores
poderá ser restabelecida em líquidos iónicos.
Foram estudados 5 intercaladores (Brometo de Etidio, Berberine,
Proflavine, Proflavine Reduzida e DAPI), com afinidades pelas bases de ADN e
cargas diferentes, em 10 líquidos iónicos distintos. Fomos capazes de mostrar
os mecanismos envolvidos nas interações entre os intercaladores e a cadeia
de ADN e, a partir destas observações, foi possível mostrar qual dos agentes
intercalantes estudados apresentou as condições mais favoráveis para uma
intercalação em meio de líquido iónico. Constatamos que não há exclusão dos
intercaladores e que eles se associam espontaneamente ao ADN em líquidos
iónicos, contradizendo as interpretações de algumas evidências experimentais
anteriores. Com base nos estudos de PMF complementado com a análise das
interações energéticas, foi possível concluir que o DAPI é o intercalador que
apresenta a intercalação mais forte e estável e, com base nos resultados de
PMF, o Liquido Iónico que promove uma intercalação mais forte e espontânea,
é o [BPYRR+] [PF6-]. Intercalating molecules are used as agents of detection and quantification of DNA. These molecules intercalate between DNA double chains in aqueous solution due to hydrophobic interactions between this molecules. These intercalating molecules are fluorescent but, when intercalated, they exhibit a significant increase in their fluorescence intensity. This effect is not observed when the solution ceases to be the usual aqueous solution but, a solution composed of an ionic liquid at room temperature (RTILs). The decrease of fluorescence of the intercalating agents, might be related to the fact that they do not intercalate within DNA helix, due to the strong ionic interactions between the solvent and DNA chain, or by other phenomena involved in the interaction between these molecules. With this project we intend to verify if this is a phenomenon of exclusion by the Ionic Liquid, or if the intercalation is spontaneous by studying the molecular mechanisms involved in the intercalation and in what condition the interaction between DNA and the intercalator may be restored in liquids ion. Five intercalators were studied (ethidium bromide, Berberine, Proflavine, reduced Proflavine and DAPI) with different DNA base-pair affinity and different loads, in ten different ionic liquids. We were able to show the mechanisms involved in the interactions between intercalators and DNA chain and, from these observations, it was possible to show which of the intercalating agents studied showed the most favorable conditions for an intercalation in ionic liquid medium. We observed that there is no exclusion of the intercalators and they spontaneously associate with DNA in ionic liquids, contradicting the interpretation of some previous experimental evidences. Based on studies of PMF, complemented with the analysis of energetic interactions, we concluded that the DAPI is the intercalator with the stronger and more stable and, based on PMF results, the ionic liquid which promotes a stronger and spontaneous intercalation is the [BPYRR+] [PF6-]. |
