Resumo(s): | A catecol-O-metiltransferase é uma enzima dependente de magnésio (Mg2+) que possui um papel primordial no metabolismo das catecolaminas, principalmente da adrenalina (AD). A baixa actividade da COMT tem sido implicada em várias condições patológicas, incluindo as relacionadas com o sistema cardiovascular. Por sua vez, tem sido sugerida uma relação inversa entre o teor de Mg2+ da água de consumo e a incidência e a mortalidade por hipertensão e doença cardiovascular. O objectivo deste estudo foi testar a hipótese de que uma relativa insuficiência do Mg2+ conduz a uma menor actividade da COMT, e uma maior ingestão de Mg2+ favorece a actividade desta enzima. O efeito destas diferentes dietas na expressão da COMT também foi avaliado. Ratos Wistar--Han adultos foram sujeitos, durante 7 semanas, a 3 diferentes dietas: grupo 1 – controlo; grupo 2 – água com alto teor em sódio (Na+) e pobre em Mg2+; grupo 3 – água com alto teor em Na+ e Mg2+. No final do tratamento, avaliou-se a expressão da COMT no fígado e na suprarrenal por rt-PCR e determinou-se a actividade da COMT, nos mesmos órgãos, por exposição à AD e ao ácido 3,4 - dihidroxifenilacético e quantificação por HPLC-ED dos metabolitos respectivos. Os animais sujeitos a uma água com maior teor de Mg2+ apresentaram maior actividade da COMT hepática (Vmax = 26,46 ± 2 nmol/h/mg; Km = 56,6 ± 18 µM) relativamente aos que foram sujeitos a uma água com alto teor em Na+ e pobre em Mg2+ (Vmax = 16,76 ± 1 nmol/h/mg; Km = 77,2 ± 26 µM), mas não foram encontradas diferenças na expressão desta enzima entre os 3 grupos. Por outro lado, na suprarrenal não foram observadas diferenças na actividade da COMT, mas a sua expressão diminuiu significativamente com o maior consumo de Mg2+, sendo 46% inferior neste grupo comparativamente ao grupo controlo (n=4-6; p<0,05). Estes resultados sugerem que o conteúdo dietético em Mg2+ pode influenciar a expressão e a actividade da COMT, sendo que este pode ser um dos mecanismos que explicam a associação entre os níveis baixos deste mineral na água de consumo e um maior risco cardiovascular. ACatechol-O-methyltransferase is a magnesium (Mg2+) dependent enzyme with a crucial role in catecholamine metabolism, mainly of adrenaline. A low COMT activity has been implicated in several health disorders, including some related with the cardiovascular system. Furthermore, an inverse relationship between the Mg2+ levels in drinking water and the incidence and mortality due to hypertension and cardiovascular disease has been suggested. The aim of this work was to test if a relative insufficiency of Mg2+ leads to a lower COMT activity, and if a higher Mg2+ intake increases the enzyme activity. The effect of these different diets on COMT expression was also evaluated. Adult Wistar-Han rats were treated for 7 weeks with 3 different diets: group 1 - control; group 2 – water with a high sodium (Na+) and low Mg2+ content; group 3 – water with a high Na+ and high Mg2+ content. At the end of the treatment, COMT expression was evaluated in the liver and adrenal glands through rt-PCR, and COMT activity was determined, in the same tissues, through exposure to adrenaline and 3,4 - dihydroxyphenylacetic acid and quantification of their respective metabolites by HPLC-ED. The animals treated with a water of higher Mg2+ content presented a higher hepatic COMT activity (Vmax = 26,46 ± 2 nmol/h/mg; Km = 56,6 ± 18 µM) than the animals treated with water with high Na+ and low Mg2+ content (Vmax = 16,76 ± 1 nmol/h/mg; Km = 77,2 ± 26 µM). There were no differences in hepatic COMT expression between the 3 groups. In the adrenal glands, no differences in COMT activity were observed, but COMT expression significantly decreased with the higher Mg2+ intake, being 46% lower than in the control group (n=4-6; p<0,05). These results suggest that dietary Mg2+ content may influence COMT expression and activity, being that this might be one of the mechanisms underlying the association between the low levels of this mineral in drinking water and a higher cardiovascular risk. |
