Resumo(s): | A angiogénese tumoral leva a um aumento da proliferação de vasos sanguíneos a
partir do tumor. Os vasos vão nutrir o tumor, promovendo o seu crescimento, mas
também vão fazer a ligação entre as células tumorais e a circulação sistémica permitindo
a disseminação destas células à distância, produzindo metástases. A importância do
VEGF (vascular endothelial growth factor) e um dos seus receptores (VEGFR-2) na
angiogénese tumoral tornaram este sistema um dos alvos mais promissores na terapia do
cancro. A inibição dos domínios de tirosina cinase do receptor aparenta ser a melhor
abordagem terapêutica.
Tendo surgido na literatura alguns estudos recentes em que derivados de
tienopiridinas mostraram actividade anti-tumoral e anti-angiogénicas, neste trabalho
sintetizaram-se alguns compostos novos com o mesmo propósito.
Obtiveram-se produtos resultantes de acoplamentos catalisados por paládio (C-C de
Suzuki-Miyaura e de C-N de Buchwald-Hartwig) e de substituições nucleofílicas
aromáticas, derivados de tieno[3,2-b]piridinas funcionalizadas, também preparadas, tanto
no anel da piridina como no do tiofeno. As tienopiridinas usadas como componentes de
acoplamento, 3-amino-6-bromotieno[3,2-b]piridina-2-carboxilato de metilo e 3-
aminotieno[3,2-b]piridina-2-carboxilato de metilo foram preparadas por reacção da 5-
bromo-3-nitropiridina-2-nitrilo ou da 3-fluoro-3-piridina-2-nitrilo com tioglicolato de
metilo em meio básico, respectivamente. 7-Halotieno[3,2-b]piridinas foram sintetizadas a
partir da 7-hidroxitieno[3,2-b]piridina comercial com POBr3 ou POCl3 tendo-se obtido a
7-bromotieno[3,2-b]piridina e a 7-clorotieno[3,2-b]piridina, respectivamente.
A partir da 7- bromotieno[3,2-b]piridina e de fluoroanilinas, obtiveram-se produtos
de acoplamento C-N de Buchwald-Hartwig, com o átomo de F nas posições orto, meta e
para. Usando a 7-clorotieno[3,2-b]piridina e aminotiofenóis prepararam-se
di(hetero)ariltioéteres com o grupo amino livre, por substituição nucleofílica aromática.
Os vários compostos aminados derivados de tieno[3,2-b]piridinas obtidos tanto por
acoplamentos catalisados por paládio nos dois anéis do sistema, como por substituição
nucleofílica no anel de piridina, reagiram com diferentes iso(tio)cianatos tendo-se obtido
as correspondentes (tio)ureias.
As constantes de inibição (Ki) das novas (tio)ureias sintetizadas foram estimadas
através de docking molecular utilizando o programa Autodock Vina no centro activo de
tirosina cinase da estrutura de Raio-X do VEGFR-2, em colaboração com o Instituto
Superior de Bragança. The tumoral angiogenesis increases blood vessels proliferation from the tumor. The vessels will nourish the tumor, promoting its growth, but they will also make the link between tumor cells and the systemic circulation, allowing cells distance dissemination, producing metastases. The importance of the VEGF (vascular endothelial growth factor) and one of their receptor (VEGFR-2) in tumor angiogenesis made this system one of the most promising targets in cancer therapy. Inhibition of this receptor tyrosine kinase domain appears to be the best therapeutic approach. Having emerged in the literature some recent studies in which derivatives of thienopyridines showed antitumoral and antiangiogenic activity, in this work some new compounds with the same purpose were synthesized. Palladium-catalyzed coupling products (C-C Suzuki-Miyaura and C-N Buchwald- Hartwig) and products resulting from nucleophilic aromatic substitution, derivatives of functionalized thieno[3,2-b]pyridines, also prepared, either in the pyridine or in the thiophene ring, were synthesized. The thienopyridines used as coupling components the methyl 3-amino-6-bromothieno[3,2-b]pyridine-2-carboxylate and the methyl 3- aminothieno[3,2-b]pyridine-2-carboxylate were prepared by reaction of the 5-bromo-3- nitropyridine-2-nitrile or of the 3-fluoro-3-pyridine-2-nitrile with methyl thioglycolate in basic medium, respectively. 7-Halothieno[3,2-b]pyridines were synthesized from the commercial 7-hydroxythieno[3,2-b]pyridine with POBr3 or POCl3 giving the 7- bromothieno[3,2-b]pyridine and the 7-chlorothieno[3,2-b]pyridine, respectively. From the 7- bromothieno[3,2-b]pyridine and fluoroanilines, Buchwald-Hartwig CN coupling products were obtained, bearing the F atom in the ortho, meta and para positions. Using the 7-chlorothieno[3,2-b]pyridine and aminothiophenols, di(hetero)arylthioethers with the free amino groups were obtained by nucleophilic aromatic substitution. The various aminated compounds derivatives of thieno[3,2-b]pyridines obtained either by palladium-catalyzed couplings in both rings of the system or by nucleophilic aromatic substitution in the pyridine ring, reacted with different iso(thio)cyanates giving the corresponding (thio)ureas. The inhibition constant (Ki) of the new synthesized (thio)ureas were estimated by molecular docking using the program Autodock Vina in the active centre of the tyrosine kinase of X-ray structure of VEGFR-2, in collaboration with the Polytechnic Institute of Bragança-Portugal. |
