Resumo(s): | O desenvolvimento de tecnologias que possibilitam a monitorização da interação de
nanomateriais com estruturas biológicas tem impulsionado a criação de novas técnicas e
dispositivos de diagnóstico e terapêutica.
A principal motivação que tem incentivado a investigação das características invulgares
das nanopartículas deve-se ao efeito quântico inerente ao tamanho (e forma) desta classe de
nanomateriais. Assim, pretende-se que a Nanotecnologia, ou seja, a exploração das
potencialidades dos materiais à nanoescala, a par da Nanociência, possibilite a criação de
nanoestruturas e fases metastáveis cujas propriedades não convencionais (incluindo a
supercondutividade e o magnetismo) possam melhorar o mundo em que vivemos.
Entre as muitas abordagens para a estruturação de nanopartículas, destaca-se o tipo coreshell
que permite evitar os efeitos indesejáveis das nanopartículas e melhorar as suas
propriedades. Os SERS Nanotags são um exemplo de materiais nanoestruturados de tipo coreshell,
que integram nanopartículas de natureza metálica e moléculas ativas de Raman (moléculas
repórter) encapsuladas com materiais biocompatíveis, como a sílica.
No âmbito desta tese, pretende-se explorar este tipo de nanoestruturas para deteção de
moléculas biológicas através da técnica de espectroscopia de difusão Raman amplificada (SERS).
Deste modo, as NPs magnéticas utilizadas, que servem como núcleo para este sistema, são de
ferrite de cálcio (CaFe2O4). A ativação da superfície destas NPs com grupos –OH, possibilita a
formação de ligações intermoleculares fortes com outras (bio)moléculas presentes no meio de
dispersão. O triptofano é a molécula repórter escolhida e a cisteína é o aminoácido que pode
estabelecer interações de afinidade com a superfície das NPs metálicas de ouro ou prata. The development of technologies that enable a monitoring of the interaction of nanomaterials with biological structures has driven the creation of new diagnostic and therapeutic techniques and devices. The main motivation that has stimulated the investigation of the unusual characteristics of nanoparticles is due to the quantum effect inherent in the size (and shape) of this class of nanomaterials. Thus, it is intended that nanotechnology, ie the exploitation of the potential of materials at the nanoscale, along with Nanoscience, will enable the creation of nanostructures and metastable phases whose unconventional properties (including superconductivity and magnetism) can improve the world in which we live. Among the many approaches to structuring nanoparticles, we highlight the core-shell type that allows to avoid the undesirable effects of nanoparticles and improve their properties. SERS Nanotags are an example of nanostructured core-shell materials, which incorporate metallic nanoparticles and active Raman molecules (reporter molecules) encapsulated with biocompatible materials such as silica. In this thesis, it is intended to explore this type of nanostructures for the detection of biological molecules through the technique of amplified Raman dispersion (SERS) spectroscopy. Thus, the magnetic NPs used, which serve as the nucleus for this system, are calcium ferrite (CaFe2O4). Activation of the surface of these NPs with - OH groups enables the formation of strong intermolecular bonds with the biological molecules present in the dispersion medium. Tryptophan is the reporter molecule chosen and cysteine is the amino acid that can establish affinity interactions with the surface of metallic gold or silver NPs. |
